Working Mechanism
Lipase functions as a serine hydrolase, adeptly catalyzing the breakdown of fatty acid glycerides into glycerol and fatty acids. The unique hydrolysis reaction occurs at the oil-water interface. Beyond this, lipase also orchestrates ester synthesis and transesterification reactions, offering a multi-faceted approach to chemical transformation.

1. Temperature range: Lipase demonstrates effectiveness between 30-70ºC,
with an optimal performance noted within the 55-65ºC range.
2. pH range: It is effective across a wide pH spectrum from 4.0-10.0,
with an ideal pH balance at 7.5 to maximize activity.
3. Impact of metal ions on activity: Calcium (Ca2+ ) and Magnesium (Mg2+ ) notably enhance enzyme action, while Manganese (Mn²+), Iron (Fe2+ ), and Zinc (Zn2+ ) can inhibit it. Meanwhile, Copper (Cu2+ ) and EDTA show minimal influence on its enzymatic prowess.

2. Enzyme activity definition: 1g of solid enzyme powder (or 1mL liquid enzyme) at specified conditions can hydrolyze substrates to yield 1 µmol of titratable fatty acids per minute, equating to 1 unit of enzyme activity (u/g or U/mL).

2. In tanning, lipase precisely targets leather fats, achieving uniform softening under gentle conditions. This enhances yield and reduces chemical necessity, minimizing skin wrinkles. Ideal for degreasing and softening, recommended usage is 0.02-0.06% based on raw material quality, with activity at 30-40°C for 30-120 minutes. Pairing with alkaline protease further optimizes results.
